RESUMO

Leptospirosis is presumably an important cause of non-malarial fever in West Africa. In this study, outpatients consulting in primary care clinics during the rainy season were tested for leptospirosis, and clinical characteristics associated with leptospirosis cases were explored. Patients with fever ≥ 39°C were recruited in nine primary health care centers in Bobo Dioulasso (Burkina Faso). Diagnosis of malaria was ruled out using a rapid diagnostic test (RDT; SD Bioline Malaria®). Leptospirosis cases were defined as patients who tested positive for Leptospira IgM (Leptocheck-WB RDT and Leptospira IgM ELISA assay, Panbio) or DNA in plasma (LipL32 polymerase chain reaction [PCR]). Among 350 patients, 202 tested positive for malaria and were excluded, and 148 met the eligibility criteria and were included. Among these, 26 subjects were considered to be leptospirosis cases: 23 tested positive for Leptospira IgM (15.5%) and three tested positive by PCR (2.2%). Headaches, abdominal symptoms, and myalgia were frequently reported without any difference between leptospirosis cases and negative cases. Cough was more frequently observed among subjects testing positive for leptospirosis (P = 0.02). Water exposure, presence of a skin injury, and walking barefoot were associated with a Leptospira-positive test. All leptospirosis cases recovered without sequelae. A significant portion of outpatients with non-malarial febrile illness during the rainy season in Burkina Faso had epidemiological factors associated with leptospirosis and tested positive for Leptospira. The favorable outcome of leptospirosis cases was reassuring; this could be due in particular to the young age of the patients.

Objectives: The objectives of this study were to determine the seroprevalence and risk factors of leptospirosis among slaughterhouse workers in Burkina Faso. Methods: We performed a cross-sectional survey of slaughterhouse workers from Ouagadougou and Bobo Dioulasso between March and April 2021. Blood was collected by venipuncture and serum samples were tested using enzyme-linked immunosorbent assay and microscopic agglutination test. Questionnaires were used to collect information from these workers on sociodemographic characteristics, work activities, knowledge of zoonosis, and risky behaviors. Results: Of the 172 subjects investigated, 28 (16.28%) were found seropositive for leptospirosis using enzyme-linked immunosorbent assay or microscopic agglutination test. The main Leptospira infecting serogroup were Mini, Autumnalis, Canicola, Copenhageni, L. mayottensis (ND), Icterohaemorrhagiae, Pyrogenes/Tarassovi (cross reaction), Panama, and Ballum. Risk factors according to multivariate analysis, included residence (P = 0.02), working at the bleeding station (P = 0.03), contact with feces and urine (P = 0.04), and the practice of agriculture outside the slaughterhouse (P = 0.05). Conclusion: These findings indicate that a significant proportion of slaughterhouse workers are being exposed to pathogenic Leptospira. Public-health interventions against leptospirosis will need to target this occupational group. Proper personal protective equipment and information about the disease should be disseminated among slaughterhouses.

Rift Valley fever virus (RVFV) is an arbovirus that causes Rift Valley fever (RVF), a zoonotic disease that mainly affects domestic and wildlife ruminants and humans. The first epidemic in North-Western and West Africa occurred in Senegal and Mauritania in 1987, two countries where RVF is now endemic. Slaughterhouse workers, farmers, herders and veterinarians are at high risk of exposure to RVF. Beyond the health threat, RVF is considered to cause major socio-economic problems, specifically in developing countries where livestock farming and trade are important economic activities. Indeed, the mortality rate linked to RVF infection can reach 95-100% in newborns and young animals. In West Africa, livestock production is a key factor for food production and for national economics. Epizootics caused by RVF can therefore have serious socio-economic consequences by impacting multisectoral economics, the psycho-social health of pastoral communities, and food security. Improving prevention strategies against RVF, including vaccination, enhancing knowledge of RVF and correcting any inappropriate behaviors by populations of endemics areas, as well as better monitoring of RVF ecological factors are effective ways to better foresee and control outbreaks of RVF and its socio-economical side-effects in countries at high risk of occurrence of the disease.

Soil bacteria spend significant periods in dormant or semi-dormant states that are interrupted by resource pulses which can lead to periods of rapid growth and intense nutrient competition. Microbial populations have evolved diverse strategies to circumvent competitive interactions and facilitate coexistence. Here, we show that nutrient use of soilborne Streptomyces is temporally partitioned during experimental resource pulses, leading to reduced niche overlap, and potential coexistence. Streptomyces grew rapidly on the majority of distinct 95 carbon sources but varied in which individual resources were utilized in the first 24 h. Only a handful of carbon sources (19 out of 95) were consistently utilized (>95% of isolates) most rapidly in the first 24 h. These consistently utilized carbon sources also generated the majority of biomass accumulated by isolates. Our results shed new light on a novel mechanism microbes may employ to alleviate competitive interactions by temporally partitioning the consumption of carbon resources. As competitive interactions have been proposed to drive the suppression of disease-causing microbes in agronomic soils, our findings may hold widespread implications for soil management for plant health.

The use of medicinal plants in the control of gastrointestinal parasitosis is a promising solution for improving the productivity of sheep flocks. In order to evaluate the anthelmintic activity of Euphorbia forskallii, in vitro bioassays were performed on three life stages of Haemonchus contortus. Five aqueous extracts concentrations namely 10 mg/mL; 5 mg/mL; 2.5 mg/mL; 1.25 mg/mL and 0.62 mg/mL were used for adult worm mortality tests. Egg hatch inhibition and L3 larval migration inhibition tests were studied at 5 mg/mL; 2.5 mg/mL; 1.25 mg/mL; 0.62 mg/mL and 0.31 mg/mL. A negative control PBS and a positive control levamisole 2.5 mg/mL were established for each test. A phytochemical screening was performed to determine the presence of some secondary metabolites. The results obtained showed the presence of total polyphenols, total flavonoids and condensed tannins within the aqueous extracts of E. forskalii. A high and significant (P < 0.05) morality rate compared to the negative control with an LC50 of 2.30 mg/mL was obtained. Inhibition of egg hatch and larval migration were high and significant (p < 0.05) compared to the negative control. There was an IC50 of 1.03 mg/mL and 0.92 mg/mL respectively for inhibition of egg hatching and L3 larval migration. The present study revealed the in vitro anthelmintic activity of E. forskalii aqueous extracts and allows us to consider in perspective complementary studies to confirm this activity.

Vitis vinifera leaves (VVL) are agro-industrial waste. In the current study, the phytochemical profile of V. vinifera leaves extracts (VVLE) of two Tunisian autochthonous accessions was determined via LC-UV-ESI/MS, and their antioxidant and hepatoprotective properties were also assessed. Mice were pretreated orally with VVLE (7.5, 15 and 30 mg/kg) for 7 days, and then received acutely and by i.p. a solution CCl4 at 12% in sunflower oil (v/v). Serum levels of hepatic markers, oxidative stress indicators in liver tissue and histological changes were assessed. LC-UV-ESI/MS analysis revealed four phenolic compounds identified in both extracts with quercetin-3-O-glucuronide being the dominant constituent (23.32 ± 1.06 vs. 10.24 ± 0.12 mg/g DM, p < 0.05 for wild and cultivated accessions, respectively). The Antioxidant activity revealed a significant difference between the genotypes. Moreover, the VVLE of the wild "Nefza-I" ecotype was the most active based on antioxidant assays. Furthermore, the results showed that pre-treatment, especially with VVLE, of the wild ecotype "Nefza-I", attenuated CCl4-induced acute liver injury in a dose-dependent manner, as demonstrated by the decrease in the activities of hepatic serum function markers. This was also evidenced by a decrease in the levels of lipoperoxidation and histological damage in the liver, as well as a restoration of antioxidant enzyme activities (SOD and catalase) and an increase in the hepatic glutathione content. Our results demonstrate that VVLE possesses protective effects on CCl4-induced liver injury. Overall, the wild ecotype "Nefza-I" extract could serve as an effective protector against CCl4-induced hepatocellular oxidative stress.

Anti-inflammatory effect of Rosa canina, Salix alba, Scrophularia nodosa and Hedera helix were studied in LPS-stimulated primary peripheral blood mononuclear cells (PBMCs) from mice (n=18) by comparing homogeneous powders of small microparticles (50-100 µm, 100-180 µm and 180-315 µm) obtained from plants via a controlled differential sieving process (CDSp) versus total plant materials obtained via hydroethanolic (HE) extraction. Further, phytochemical composition of the fine powders and HE extracts was determined by LC-PDA-ESI/MS analyses. Results showed that a one-hour pretreatment of PBMCs with fine powders, particularly those with superfine particle sizes (i.e. 50-100 µm and 100-180 µm), significantly inhibited TNFα, IL-1ß, IL-6 and NO production in LPS-stimulated PBMCs, by at least ca. 20% more than HE extracts (all, p<0.05). For each of the plants studied, their superfine powdered fractions were more concentrated in phenolic contents than their HE extracts. Overall, our results further confirm CDSp, as an environmentally friendly method, for improving the concentration of bioactive compounds as well as their biological activities.

Dengue is the most widespread arbovirosis in the world, with approximately 390 million cases per year, 96 millions of which have clinical manifestations and 25,000 deaths. In West Africa, the circulation of this virus in human populations was first reported in the 1960s in Nigeria. Clinical diagnosis of dengue in West Africa is made difficult by the existence of other diseases with similar clinical presentations. Biological diagnosis remains therefore the only alternative. This biological diagnosis requires high quality equipment and well-trained personnel, which are not always available in resource-limited countries. Thus, many cases of dengue fever are consistently reported as malaria, leading to mismanagement, which can have serious consequences on the health status of patients. It is therefore necessary to set up surveillance systems for febrile infections of unknown origin in Africa by strengthening the diagnostic capacities of national laboratories.

ABSTRACT: La dengue est l'arbovirose la plus répandue dans le monde avec environ 390 millions de cas par an, dont 96 millions présentent des manifestations cliniques, avec plus de 25 152 décès annuels répertoriés. Le diagnostic clinique de la dengue en Afrique de l'Ouest est rendu difficile par l'existence d'autres maladies présentant des tableaux cliniques similaires. Il est donc nécessaire de mettre en place des systèmes de surveillance des infections fébriles d'origine inconnue en Afrique, en renforçant les capacités diagnostiques des laboratoires nationaux.

Bovine tuberculosis (bTB) is a zoonotic, infectious, chronic and contagious disease, caused by Mycobacterium bovis that mainly affects cattle. This pathology has a negative impact on animals and animal products trade. Unfortunately, in Burkina Faso where agriculture and livestock sectors represent around 80% of the socio-economic activities, the real situation of the disease is not well known especially in small ruminants and swine. Thus, our study focused on both the epidemiology and the microbiological diagnosis of tuberculosis (TB) in small ruminants and pigs slaughtered at Bobo-Dioulasso abattoir. A prospective study was conducted between August 2017 and December 2017. Epidemiological data collection was performed during routine meat inspection; moreover, samples were taken and transported to the Bacteriology laboratory of Centre Muraz for microbiological analyses. This diagnosis consisted in search of Acid Fast Bacilli (AFB) using the hot Ziehl-Neelsen staining. Out of a total of 14 648 small ruminants and 2430 pigs slaughtered during the study period, 156 and 17 had lesions suggestive of bTB with prevalence of 1.07% and 0.7%, respectively. Females and those between 2 and 4 years old were mainly infected. The most affected organs were: lungs, liver, spleen and lymph nodes. Finally, microscopy revealed 43.35% (75/173) of positive cases for AFB. These results confirm the presence of bTB in small ruminants and pigs in Burkina Faso. Efforts must still be made in the fight against this zoonosis in order to limit its economic and public health impacts.

Emerging diseases are a major public health problem as illustrated by the current coronavirus disease (COVID-19) pandemic. To make the right decisions, public health departments need a decision-making system. In Africa few IT systems have been put in place to help managers of public health in the analysis of their multidisciplinary data. The majority of digital health solutions are operational databases, as well, focused on surveillance activities that do not include the laboratory component. This paper describes the design model and implementation of data warehouse for dangerous pathogen monitoring in a laboratories network. Talend data integration is used to extract data in Excel sheets, transform it and load it into a MySQL database.

The analysis of Mycobacterial Interpersed Repetitive Unit-Variable Number of Tandem Repeat (MIRU-VNTR) discriminates against the species of M. tuberculosis involved in the transmission of the disease. The reference method is the manual method. Our study involved developing a bioinformatics method of interpreting MIRU-VNTR and comparing it to the manual method. For this we used two softwares, namely imagej and Microsoft Excel. Imagej was used to determine the migration distance of the bands and for the measure of size in a base pair. The number of repetitions of 18 markers used was analyzed with Excel macro. The results obtained were: 27% of the results exactly consistent, 16% of outliers generated by the macro and 57% of the results not matching.

This study reports the link between larvicidal activity and the phytochemical composition of male and female leaf extracts of Mercurialis annua L. (M. annua) from four Tunisian regions: Bizerte, Jandouba, Nabeul and Beja. Their antioxidant activity was evaluated using DPPH (2, 2-diphenyl-1-picrylhydrazyl) assays. Phenolic compounds were identified and quantified using liquid chromatography coupled with a UV detector and mass spectrometry (LC-UV-ESI/MS). Higher antioxidant activity (AOA) was found in the leaves of male plant extracts than of female ones. The leaves of male and female plant extracts from Bizerte exhibited the highest AOA: 22.04 and 22.78 mg Trolox equivalent/g dry matter (mg TE/g DM), respectively. For both sexes, plant extracts from Beja had the lowest AOA with 19.71 and 19.67 mg TE/g DM for male and female plants, respectively. Some phenolic compounds such as narcissin, gallocatechin, rutin, epigallocatechin and epicatechin were identified and quantified using LC-MS, which highlighted the abundance of narcissin and rutin in the male leaves of M. annua. We noted that the interaction between the sex of plants and the provenance had a significant effect on TFC (F = 6.63; p = 0.004) and AOA (F = 6.53; p = 0.004) assays, but there was no interaction between sex and origins for TPC (F = 1.76; p = 0.19). The larvicidal activity of aqueous leaf extracts of M. annua against Tribolium confusum (Du Val) (T. confusum), an insect pest of flour and cereal seeds, showed that the mortality could reach 100% after 48 h of exposure in the Bizerte region. The LC50 values for the leaf extract were low in Bizerte, with 0.003 and 0.009 g/mL for male and female plants, respectively, succeeded by Jandouba, which displayed 0.006 and 0.024 g/mL for male and female plants, respectively. Nabeul showed 0.025 g/mL for male plants and 0.046 g/mL for female plants and Beja showed 0.037 and 0.072 g/mL for male and female plants, respectively. This is the first time that a study has revealed a negative correlation between the antioxidant activity and the larvicidal activity of the leaf extracts of M. annua with the following correlation coefficients of Perason: r = -0.975 and r = -0.760 for male and female plants, respectively.

An assessment of the quality of the information system was conducted in the 14 hospitals of Burkina Faso in 2017 using the HMN tool (Health Metrics Network). This evaluation was part of the process of developing a master plan for implementing a HIS for the hospitals in the country, and was aimed at analyzing the sub-components of the existing information system. The results are presented as scores, one per component, evaluated in % and converted into quartiles, ranging from the 1st quartile "Not adequate at all" to the 4th quartile "Highly adequate". The scores are as follows: Resources = Q1 (29%); indicator = Q2 (46%); data sources = Q1 (28%); information production = Q1 (21%); data management = Q2 (47%); dissemination and use = Q3 (55%). In conclusion, overall the system is not adequate (mean score 39%), particularly at the level of information production (score 21%).

INTRODUCTION: Fennel (Foeniculum vulgare Mill.) and basil (Ocimum basilicum L.) are two aromatic medicinal plants. Recently, a new parting process which is named CDS "Comminution and to control Diffraction Sieving" is taken into consideration and its positive effect on the extraction of bioactive compounds from the plants, without any solvent, is reported. OBJECTIVE: Study the effect of CDS on phytochemical properties of superfine powders of fennel seeds and basil leaves. METHODS: Fennel seeds and basil leaves superfine powders were fractionated as follows: 100-180 µm, 180-315 µm, 315-500 µm, > 500 µm and unsieved superfine powders. Extraction of polyphenols was carried out using hydromethanolic maceration. The essential oils were extracted by maceration with dichloromethane. The antioxidant activities were evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. Using liquid chromatography ultraviolet electrospray ionisation mass spectrometry (LC-UV-ESI-MS), concentrations of characterised phenolic compounds were measured. The essential oils compositions were characterised using gas chromatography mass spectrometry (GC-MS). RESULTS: In fennel seeds, the 100-180 µm fraction had maximum antioxidant activity and LC-UV-ESI-MS analyses proved that the best extraction of certain polyphenols was obtained in the 100-180 µm fraction. In basil leaves, the best antioxidant activity corresponded to the 315-500 µm fraction and LC-UV-ESI-MS analyses showed that the polyphenols were concentrated in the < 315 µm fractions. For both plants, GC-MS presented that the essential oils were concentrated in the 315-500 µm fraction. CONCLUSION: CDS as a new green parting process leads to improve the phytochemical properties of these two superfine plant powders in the specific granulometric classes.

AIM: The study aimed to evaluate the occurrence of anti-Toxoplasma gondii and anti-Neospora caninum antibodies in sheep breeding in a particular husbandry system called "Elevage en case" in Dakar, Senegal. MATERIALS AND METHODS: Blood samples were collected from 278 sheep. Serum was harvested and used for analysis. For the detection of T. gondii antibodies, 278 sera were analyzed using the modified agglutination test, while the enzyme linked-immunosorbent assay was used on 174 sheep sera to look for N. caninum antibodies. RESULTS: This study showed that 29.4±6.8% of sheep carried both T. gondii and N. caninum antibodies. The overall prevalence was 60.1±5.7% and 41.9±7.3% for toxoplasmosis and neosporosis, respectively. For toxoplasmosis, the prevalence was higher in Gueule Tapée (63.3%) than in Medina (58.9%), but the variation was not significant (p=0.45). Regarding the age of animals, the prevalence was significantly higher in animals over 2 years old compared to those under 2 years old (p=0.002). For neosporosis, the prevalence was significantly higher in Medina (48.67%) than Gueule Tapée (16.7%) (p=0.001), but non-significant variation was noted according to animal age. CONCLUSION: The study showed that sheep reared in households have carried antibodies of T. gondii and N. caninum. The prevalence was high and it means that consumption of meat from these animals is risky if the meat is eaten undercooked.

Hepatitis E virus infection is a significant public health problem in many parts of the world including Africa. We tested serum samples from 900 patients in Burkina Faso presenting with febrile icterus. They all tested negative for yellow fever, but those from 23/900 (2.6%) patients contained markers of acute HEV infection (anti-HEV IgM and HEV RNA positive). Genotyping indicated that 14 of the strains were HEV genotype 2b. There was an overall HEV IgG seroprevalence of 18.2% (164/900). In a bivariate analysis, the factors linked to HEV exposure were climate and patient age. Older patients and those living in arid regions were more likely to have HEV infection. HEV genotype 2b circulating only in humans can be involved in some acute febrile icterus cases in Burkina Faso. Better access to safe water, sanitation, and improved personal hygiene should improve control of HEV infection in this country.

In West Africa, identification of nonmalarial acute febrile illness (AFI) etiologic pathogens is challenging, given limited epidemiologic surveillance and laboratory testing, including for AFI caused by arboviruses. Consequently, public health action to prevent, detect, and respond to outbreaks is constrained, as experienced during dengue outbreaks in several African countries. We describe the successful implementation of laboratory-based arbovirus sentinel surveillance during a dengue outbreak in Burkina Faso during fall 2017. We describe implementation, surveillance methods, and associated costs of enhanced surveillance during an outbreak response as an effort to build capacity to better understand the burden of disease caused by arboviruses in Burkina Faso. The system improved on existing routine surveillance through an improved case report form, systematic testing of specimens, and linking patient information with laboratory results through a data management system. Lessons learned will improve arbovirus surveillance in Burkina Faso and will contribute to enhancing global health security in the region. Elements critical to the success of this intervention include responding to a specific and urgent request by the government of Burkina Faso and building on existing systems and infrastructure already supported by CDC's global health security program.

This work evaluated the effect of grinding and sieving process of green tea leaves on particle size distribution and antioxidant activity. Granulometric classes ranging from 20 µm to 500 µm were studied. Hydroalcoholic extracts of green tea powders (GTPs) were analysed for total phenolic, total flavonoid, and catechin contents. The fraction of 100-180 µm ground at 6000 rpm showed the maximum catechin content with 33.5 mg/g dry matter (DM) and, the best antioxidant activity with IC50 values of 0.28 µg/mL and 0.13 µg/mL using DPPH and ABTS radical scavenging assays, respectively. However, antioxidant properties and catechin content decreased for particle of sizes less than 50 µm. Catechin content was higher for those ground at 6000 rpm as compared to at 18,000 rpm. The best grinding conditions and particle size were 6000 rpm and 100-180 µm.

Phytoremediation is considered as a promising and cost-effective method to enhance bioremediation of polluted soils. Exudation of plant root secondary metabolites similar to organic pollutants may induce the expression of microbial degradative enzymes and favour cometabolism of xenobiotics. We investigated the contribution of Miscanthus x giganteus root exudates in the biostimulation of PAH-degradation. This perennial grass was chosen because of its capability to grow on polluted soils and its high biomass production for non-food purposes. First, the impact of cometabolism phenomena was evaluated on the selective enrichment of pyrene-degrading bacterial consortia. The identification of each isolated strains following incubation with pyrene only, "pyrene+phenanthrene", "pyrene+salycilate" or "pyrene+diesel fuel" showed a varying bacterial diversity and pyrene-degrading ability, depending on the co-substrate used. Then, a microplate assay was designed, based on the simultaneous measurement of bacterial consortia growth and degradation activity, in the presence of PAH and total root exudates. Results showed that i) the addition of root exudates was efficient for promoting bacterial growth, ii) but a selective enrichment of PAH-degraders compared to aliphatic ones could be clearly demonstrated, thereby conducing to an enhanced PAH catabolism. The identification of plant secondary metabolites showed the presence of a broad range of flavonoid-derived compounds that could play a role in cometabolic processes. Microplate assays with the two major molecules, quercetin and rutin, suggested a partial involvement of these compounds in biostimulation processes. Further investigations with the other identified secondary metabolites (apigenin, isovitexin, catechin, gallic and caffeic acid) should provide more information on the exudate-PAH cometabolic degradation phenomenon.

Desmodium adscendens plant is widely used as juice or tea in various parts of the world against a wide range of diseases. This study determines the quality and the quantity of polyphenols, flavonoids, anthocyanins, and tannins in D. adscendens leaves by UV-spectrophotometry and RP-HPLC methods. In addition, the antioxidant capacity of these phenolic compounds is evaluated by ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic)), DPPH (2,2-diphenyl-1 picrylhydrazyl), and Cellular tests. D. adscendens leaves are mainly composite of flavonoid compounds with 12.8 mg of catechin equivalent (CE)/g dw. The amounts of total polyphenol compounds are 11.1 mg of gallic acid equivalent (GAE)/g dw. The quantity of total anthocyanin and total tannin compounds is not considerable 0.0182 mg CgE/g dw and 0.39 mg CE/g dw, respectively. A direct correlation between phenolic compounds and antioxidant activity is observed (R(2) = 0.96). The RP-HPLC analyses reveal that the main phenolic compound identified in the methanol-water extract is quercetrin dihydrat (2.11 mg/mL). According to the results, it is observed that D. adscendens leaves possess a considerable scavenging antioxidant and antiradical capacity, therefore these antioxidant properties might increase the therapeutic value of this medicinal plant.
